Why is Ademetionine Prescribed? (Indications)

Ademetionine is commonly prescribed for:

- Chronic Liver Disease: Including conditions like intrahepatic cholestasis, where bile flow is reduced, leading to accumulation.

- Mood Disorders: Such as depression.

- Osteoarthritis: To alleviate joint pain.

- Muscle Pain/Fibromyalgia: To manage pain and associated symptoms.

Contraindications: When Should Ademetionine Not Be Taken?

- Bipolar Depression: Ademetionine is contraindicated as it may increase anxiety and mania.

- Allergic Reactions: It should not be used by individuals allergic to the drug.

 Dosage Guidelines for Ademetionine

Depression:

- Oral: 400-1600 mg per day

- IM/IV: 200-400 mg/day

Osteoarthritis:

- Oral: 200 mg three times daily

- IV: 400 mg/day

Liver Disease (Alcohol-Related):

- Oral: 1200-1600 mg per day; not recommended for use beyond 2 years

- IV: 200-2000 mg/day for 15-30 days

- IM: 200 mg/day for 30 days

Intrahepatic Cholestasis:

- Oral: 800 mg twice a day

Fibromyalgia:

- Oral: 800 mg per day

Administration Instructions

- Tablets: Should be swallowed whole.

- Timing: Best taken on an empty stomach for optimal absorption and therapeutic effect.

 Warnings and Precautions

- Kidney Disease: Caution is advised.

- Pregnancy: Avoid during the first trimester unless advised by a healthcare provider.

- Breastfeeding: Should be avoided.

- Children: Not recommended for use in children.

- Pre-surgery: Should be discontinued at least two weeks before surgery.

- Vitamin Deficiencies: Patients with folate and vitamin B12 deficiencies should monitor their levels regularly.

- Liver Disease: Ammonia levels should be monitored.

 Potential Side Effects

- Gastrointestinal: Vomiting, gas, constipation, diarrhea, loss of appetite, dry mouth.

- Nervous System: Headache, insomnia, dizziness, nervousness.

- Serotonin Syndrome: Symptoms include confusion, anxiety, tremors, diarrhea, high blood pressure, and seizures.

- Other: Sweating, skin rashes, allergic reactions, including anaphylaxis.

 Drug Interactions

- Antidepressants: Increases risk of serotonin syndrome when combined with drugs like fluoxetine, sertraline, clomipramine, and supplements like St. John's wort.

- Antidiabetes Medication: Can cause severe reductions in blood sugar levels.

- Temperature: Store at temperatures not exceeding 30°C.
